Functions of Solvents

  • Vehicle for Liquid Preparations: Solvents provide the bulk medium for liquid dosage forms such as oral syrups, injectables, ophthalmic drops, and topical solutions. The solvent not only maintains consistency and uniformity but also affects taste, texture, and patient acceptability.
  • Processing Aids in Manufacturing: In pharmaceutical production, solvents assist in wet granulation, purification and crystallization of APIs, extraction of natural actives, polymer coating of tablets, and preparation of gels and suspensions.
  • Enhancing Stability: Certain solvents can stabilize sensitive APIs by preventing oxidation, hydrolysis, or microbial growth. For example, glycerin and propylene glycol possess inherent antimicrobial and humectant properties, adding functional value to the formulation.
  • Controlled Drug Release: In advanced drug-delivery systems (DDS), solvents help control polymer behavior, drug diffusion rates, and solid-state transitions. They are especially important in sustained-release injectables and transdermal systems.

Types of Common Solvent Excipients

Water

Water, especially purified water and water for injection (WFI), is the primary solvent used in pharmaceuticals due to its safety and broad compatibility. Purified water is suitable for oral and topical products, while WFI meets stringent microbial limits required for injectables.

Alcohols

Alcohols such as ethanol and isopropyl alcohol serve as effective solvents for hydrophobic or semi-polar active ingredients, playing an important role in both formulation and manufacturing. Ethanol is commonly used in oral solutions, tinctures, and topical preparations due to its strong solubilization ability and antimicrobial properties, while isopropyl alcohol is primarily reserved for external applications, disinfection, and processing steps.

Polyols

Polyols, including propylene glycol (PG), glycerin, and various grades of polyethylene glycol (PEG), are versatile pharmaceutical solvents valued for their low toxicity, strong solubilizing ability, and excellent safety record. These semi-polar solvents are widely used in oral liquids, injectables, topical gels, and suppositories, where they improve viscosity, stabilize sensitive APIs, and act as humectants.

Esters

Ester solvents such as ethyl acetate and butyl acetate are primarily processing solvents used in coating, crystallization, and purification. Their favorable solvency and rapid evaporation support efficient tablet-coating operations and help achieve desired crystal forms of APIs. Although not commonly used directly in finished dosage forms, they play an important role in manufacturing steps that influence product quality.

Hydrocarbons

Hydrocarbons like hexane, heptane, and mineral oil are applied in extraction, and specific formulation needs. Hexane and heptane assist in API purification and crystallization due to their low polarity, while mineral oil serves as both solvent and emollient in topical preparations. Strict residual limits apply, but hydrocarbons remain valuable in processes that require selective solvation or moisturizing functionality.

Ethers

Ethers such as propylene glycol monomethyl ether (PM) and diethylene glycol monoethyl ether provide strong solubilizing capability and excellent membrane penetration, making them suitable for transdermal, dermal, and some injectable applications. Their compatibility with various APIs and polymers supports improved bioavailability and flexible formulation design. Ethers are increasingly used in advanced dosage forms requiring efficient drug delivery through the skin or tissues.
